چکیده

The reaction between hemoglobin (Hb) and NO • has been investigated thoroughly in the last years but its mechanism is still a matter of substantial controversy. We have carried out a systematic study of the influence of the following factors on the yield of S-nitrosohemoglobin (SNO-Hb) generated from the reaction of NO • with oxy-, deoxy-, and metHb: 1) the volumetric ratio of the protein and the NO • solutions; 2) the rate of addition of the NO • solution to the protein solution; 3) the amount of NO • added; and 4) the concentration of the phosphate buffer. Our results suggest that the highest SNO-Hb yields are mostly obtained by very slow addition of substoichiometric amounts of NO • from a diluted solution. Possible pathways of SNO-Hb formation from the reaction of NO • with oxy-, deoxy-, and metHb are described. Our data strongly suggest that, because of mixing artefacts, care should be taken to use results from in vitro experiments to draw conclusion on the mechanism of the reaction in vivo.

Reactions of deoxy-, oxy-, and methemoglobin with nitrogen monoxide. Mechanistic studies of the S-nitrosothiol formation under different mixing conditions.

A tri state mechanism for oxygen release in fish hemoglobin: Using Barbus sharpeyi as a model

Hemoglobin is a porphyrin containing protein with an a2b2 tetrameric structure and like other porphyrin compounds shows spectral behavior of species specific characteristics. Researchers tend to relate bands in the hemoglobin spectra to certain structural and/or functional features. Given the fact that hemoglobin is the main oxygen carrier in animals functioning through the Oxy«Deoxy equilibriu...

Characterization of Sol-Gel Derived CuO@SiO2 Nano Catalysts towards Gas Phase Reactions

One distinct concentration of copper ions was embedded into the silica matrix to xerogel form using copper source Cu(NO3)2∙3H2O. The xerogel samples were prepared with using hydrolysis and condensation reactions of TetraEthyl Ortho-Silicate (TEOS) by the sol-gel method. In this investigation, new molar ratio of H2</su...

Antioxidant effect of different vitamins on methemoglobin production: An in vitro study

Nitrite intoxication occurs frequently in ruminants and equines. The most common treatment of this disorder is administration of 1% methylene blue, although the use of some antioxidant agents e.g. vitamins and complementary treatment may also be useful. The aim of this study was to evaluate the in vitro antioxidative effects of some vitamins on methemoglobinemia induced by sodium nitrite. For t...

Aggregation and crystallization of hemoglobins A, S, and C. Probable formation of different nuclei for gelation and crystallization.

The oxy and carbonmonoxy forms of Hb A and Hb S formed aggregates or gels when dissolved in phosphate buffers at concentrations above their solubility and warmed rapidly to 30 degrees C from 0 degrees C. Kinetic studies showed that although deoxy-Hb A and deoxy-Hb S aggregated with a clear exhibition of a delay time, the oxy and carbonmonoxy forms of Hb A and Hb S did not show a delay time. The...
